소포체 스트레스 (ER stress) and Unfoled Protein Response (UPRER)
소포체는 세포내 소기관으로 분비 또는 막단백질의 Folding이 일어나는 곳이며, 세포내 칼슘의 항상성 조절, 그리고 세포 막지질의 생합성이 일어나는 곳임 (The endoplasmic reticulum (ER) is an organelle within the cell where secretion and the folding of membrane proteins occur. It plays a crucial role in regulating the homeostasis of cellular calcium and in the biosynthesis of cellular membrane lipids)
소포체의 항상성이 교란될 경우 세 가지 신호전달경로가 활성화되는데 이는 PERK, IRE1a, 그리고 ATF6a라는 막단백질에 의해 매개됨 (When the homeostasis of the endoplasmic reticulum (ER) is disrupted, three signaling pathways are activated. These pathways are mediated by membrane proteins called PERK, IRE1α, and ATF6α. )
각각 ATF4, XBP1s 및 ATF6a라는 세 종류의 전사조절인자의 발현이 유도되며, 이들에 의해 하위 유전자들의 전사가 증가하게 됨 (In response to ER stress, the expression of three transcription factors, ATF4, XBP1s, and ATF6α, is induced. 소포체 스트레스는 다양한 세포 내외의 자극에 의해 발생하며, 이를 극복하기 위한 UPR이 유도되지만, 스트레스가 해소되지 못할 경우 결국 세포의 기능 이상 및 죽음으로 연결되며, 이는 최종적으로 인간질병으로 발전하게 됨 (Endoplasmic reticulum (ER) stress can be triggered by various intracellular and extracellular stimuli. When ER stress occurs, the Unfolded Protein Response (UPR) is induced to restore ER homeostasis and promote cell survival. However, if the stress is not alleviated, it can lead to dysfunction and ultimately cell death. Prolonged or severe ER stress has been implicated in the development and progression of various human diseases, including neurodegenerative disorders, metabolic diseases, and inflammatory conditions. Understanding and modulating the UPR pathway have become important targets for therapeutic interventions in these diseases. )

미토콘드리아 스트레스 (Mitochondrial stress) and Unfoled Protein Response (UPRMT)
미토콘드리아는 세포가 살아가는데 필요한 에너지를 ATP의 형태로 생성하는 중요한 역할을 하는 것 뿐 아니라, 칼슘의 항상성, 활성산소 제어, 세포자멸사 (Apoptosis) 제어 및 다양한 신호전달경로 제어 등의 다양한 기능을 수행하는 중요한 세포 소기관임 (Mitochondria, as essential organelles, not only play a crucial role in generating the energy required for cell survival in the form of ATP (adenosine triphosphate), but they also perform various other functions including calcium homeostasis, regulating reactive oxygen species (ROS), controlling apoptosis (programmed cell death), and modulating various signaling pathways)
미토콘드리아가 올바르게 작동을 하기 위해서는 미토콘드리아의 다양한 기능에 관여하는 단백질들의 항상성이 유지되어야 하며, 이러한 단백질 항상성이 교란되면 단백질들의 기능 손상으로 이어져 결과적으로 미토콘드리아의 기능이 상실됨. 따라서 이러한 단백질 항상성 교란을 원활히 복구하는 것은 미토콘드리아의 기능성에 있어서 매우 중요하며, 이러한 복구 시스템을 우리는 미토콘드리아 URP (Mitochondrial Unfolded Protein Response, UPRMT)라 칭함 (For mitochondria to function properly, the protein homeostasis involved in various mitochondrial functions must be maintained, as disruption of protein homeostasis leads to protein dysfunction and, ultimately, mitochondrial dysfunction. Therefore, the efficient restoration of protein homeostasis is crucial for mitochondrial functionality, and this recovery system is referred to as the Mitochondrial Unfolded Protein Response (UPRMT))
C.elgans에서의 UPRMT의 경우는 매우 잘 알려져 있는 반면, 포유동물에서의 UPRMT는 그 기전이 명확하지 않음. 최근 미토콘드리아 스트레스 상황에서 DELE라는 단백질의 분해가 일어나며 짧은 크기의 DELE가 eIF2a 인산화 효소인 HRI를 활성화시키면서 UPRMT를 유도한다는 논문이 발표되었음. 현재 우리 실험실에서는 미토콘드리아 스트레스가 발생했을 때 double strand mitochonrial RNA (mt dsRNA)가 축적되며, 이들이 PKR을 활성화 시키면서 UPRMT를 유도한다는 사실을 관찰하였음 (The mechanism of UPRMT (Unfolded Protein Response in Mitochondria) is well known in C.elegans, but in mammals, it remains unclear. Recently, a study was published suggesting that in situations of mitochondrial stress, the protein DELE undergoes degradation, resulting in the activation of HRI, a phosphorylating enzyme of eIF2a. In our current laboratory experiments, we have observed that double-strand mitochondrial RNA (mt dsRNA) accumulates during mitochondrial stress, and they activate PKR, thereby inducing UPRMT)
미토콘드리아는 세포의 가장 중요한 소기관으로서 이들의 기능이 정상적이지 못할 경우 세포의 기능이상 또는 죽음으로 이어지며, 결국 그 세포가 이루고 있는 조직 도는 장기의 기능이상 및 관련 질병을 초래하게 됨. 따라서 미토콘드리아가 겪고 있는 스트레스를 적절히 해소하는 것이 매우 중요하며, 이러한 스트레스 반응이 적절하지 못할 경우 다양한 인간질병이 생기게 됨. 이들 질병은 대사질환, 퇴행성뇌질환, 대사염증, 근육질환, 간질환 등 매우 다양하게 포함되어 있음. 본 연구실은 미토콘드리아 스트레스에 대한 반응기전을 연구하고 있으며, 이러한 발견을 인간 질병을 이해하기 위한 접근방법에 접목시키고 있음 (Mitochondria are vital organelles within cells, and when their function is compromised, it can lead to cellular dysfunction or even cell death. Ultimately, this can result in tissue or organ dysfunction and related diseases. Therefore, it is crucial to effectively alleviate the stress experienced by mitochondria. If the stress response is inadequate, various human diseases can arise. These diseases encompass a wide range of conditions, including metabolic disorders, neurodegenerative diseases, metabolic inflammation, muscular disorders, and liver diseases, among others. Our research laboratory is studying the mechanisms of stress response in mitochondria and integrating these findings into approaches aimed at understanding human diseases)
